The gravel should be in the tank with the fish and filtration… unless the “another tank” refers to another fish stocked tank. The nitrifying bacteria needs the ammonia from the fish and decaying detritus as the starting food for the nitrogen cycle.
Technically, used gravel right out of a running tank and then put in another tank (even without fish as long as there is water circulation) will continue to cycle for a while as any of the detritus transferred with the dirty/used gravel will continue to break down putting out ammonia… perpetuating the nitrogen cycle. But if your Q-tank isn’t cycled, then putting the gravel in there will help the nitrogen cycle in your Q-tank.
The aquarium silicone cure rate would depend on how thick of a layer. When you can’t smell the vinegar-ish smell and it’s no longer putting out any odor, then it’s cured. Usually 48-72 hours… sometimes less, sometimes more.
